The stability challenge is complex, as each vitamin has a distinct degradation mechanism and the combined system must be tested with Stability Indicating methods for each active compound.
• Calcium Effervescent tablets containing Calcium Carbonate, Calcium Citrate, or Calcium Lactate are targeted at women and older adults requiring calcium supplementation. These formulations tend to be higher in tablet weight and require assessment of dissolution completeness and calcium content stability throughout shelf life.
• Electrolyte Effervescent tablets containing Sodium, Potassium, Magnesium, and Chloride are used for electrolyte replenishment after exercise or heavy perspiration. Effervescent tablets are the most moisture-sensitive format among solid dosage forms. Seven challenges specific to this format are addressed in TIBD’s stability protocol.

1. The first is extreme moisture sensitivity and Pre-reaction risk. The effervescent reaction is driven by the acid-base system: Citric Acid, Tartaric Acid, or Malic Acid reacts with Carbonate Salts (Sodium Bicarbonate, Potassium Bicarbonate) in water, releasing CO2. In packaging with any moisture ingress, Pre-reaction can begin inside the sachet or tube, consuming the effervescent system before use. The Critical Moisture Content for effervescent tablets is approximately 0.5 to 1.0%. Above this level, Pre-reaction risk increases significantly. Controlling moisture during manufacturing and storage is the most critical quality parameter for this dosage form.
2. The second is vitamin degradation. Vitamin C degrades through oxidation and is accelerated by oxygen exposure and low pH in the reconstituted solution. Vitamin B-complex compounds degrade by heat and light, with Riboflavin being particularly light-sensitive and Thiamine being heat-sensitive.
3. The third is herbal active degradation. Curcuminoids in Turmeric effervescent tablets degrade at low pH in the dissolved system and are susceptible to alkaline hydrolysis. Andrographolide in Andrographis formulations is heat-sensitive. These stability characteristics must be confirmed by Stability Indicating HPLC methods that can distinguish the marker compound from its degradation products.
4. The fourth is colour change from Maillard reaction between reducing sugars and amino acids present in some formulations, or from degradation of natural colorants such as Curcuminoids, Anthocyanins, and Carotenoids. Colour change is a first-notice consumer quality signal and is assessed by CIELAB colorimetry throughout the stability study.
5. The fifth is taste and odour change. Effervescent tablets are consumed as a beverage, making sensory attributes critical consumer quality parameters. Changes in sourness profile, sweetness intensity, or development of off-odours are assessed at each Pull Point.
6. The sixth is degradation of excipients, including Sweeteners such as Aspartame, which is heat-sensitive and can degrade to Diketopiperazine and Phenylalanine at elevated temperatures, and Flavoring Agents which may volatilize or chemically degrade over long-term storage.
7. The seventh is coating or film degradation. Some effervescent tablets use a coating or film system to protect certain actives or to delay release of specific components. Coating integrity must be assessed at each Pull Point, as cracking or delamination of the coating compromises the stability of the protected system.

• Moisture Content is measured by Karl Fischer Titration using a high-sensitivity coulometric or volumetric instrument. The specification for effervescent tablets is typically Not More Than 0.5 to 1.0%. Values exceeding this limit indicate a packaging barrier failure or inadequate humidity control during manufacturing, and are an immediate indicator of Pre-reaction risk.
• Water Activity (aw) is measured by a Water Activity Meter. The specification is typically Not More Than 0.30 for effervescent formulations. Water Activity is a more sensitive predictor of chemical stability and microbial risk than Moisture Content alone, as it reflects the availability of water for chemical reactions rather than total water content.
• Effervescence Time is measured from the moment the tablet is dropped into water at 15 to 25°C until complete dissolution, per USP <2040> or Ph.Eur. 2.9.1. The standard specification for most effervescent tablets is Not More Than 5 minutes. An increase in dissolution time indicates loss of effervescent performance from Pre-reaction or degradation of the acid-base system.
• Disintegration Testing is conducted in water at 15 to 25°C per Ph.Eur. 2.9.1. The tablet must fully dissolve and disperse within the specified time with no residual solid fragments.
• pH of the reconstituted solution is measured by pH Meter after complete dissolution. The typical specification range is pH 4.0 to 7.0 depending on the formulation. A change of more than plus or minus 0.5 pH units from the initial value indicates degradation of the acid-base equilibrium system.
• Hardness is measured by a Tablet Hardness Tester per Ph.Eur. 2.9.8. The typical specification range for effervescent tablets is 60 to 150 N. Hardness that is too high may slow the effervescent reaction; hardness that is too low may lead to tablet breakage during transport and handling.
• Friability is measured by a Friability Tester per Ph.Eur. 2.9.7. The specification is typically Not More Than 1.0% mass loss.
• Weight Variation and Content Uniformity are assessed per Ph.Eur. 2.9.5 and 2.9.6, confirming that each tablet contains the stated quantity of active ingredient within acceptable variation limits.
• Carbon Dioxide Content released during effervescence is measured by Gravimetric Method or Acid-Base Titration, confirming that the effervescent system retains sufficient CO2-generating capacity throughout the declared shelf life.

• Vitamin C (L-Ascorbic Acid): Analyzed by HPLC with UV detection at 245 nm using a Stability Indicating Method that distinguishes Ascorbic Acid from Dehydroascorbic Acid and further degradation products. The mobile phase must contain Metaphosphoric Acid to prevent oxidation of the analyte during sample preparation and analysis.

• Vitamin D3 (Cholecalciferol): Analyzed by HPLC with UV detection at 265 nm. Due to the low concentration of Vitamin D3 in most formulations (typically 400 to 2,000 IU per tablet), a concentration step may be required.
• Calcium Content: Measured by Atomic Absorption Spectroscopy (AAS) or Inductively Coupled Plasma Optical Emission Spectrometry (ICP-OES), which provides accurate measurement of calcium in the dissolved matrix.
• Magnesium, Zinc, Iron, and Other Minerals: Measured by AAS or ICP-MS depending on the concentration level and the complexity of the mineral matrix.
• Curcuminoids in Turmeric Effervescent: Analyzed by HPLC with UV detection at 425 nm using a Stability Indicating Method that separates Curcumin, Demethoxycurcumin, and Bisdemethoxycurcumin. • Andrographolide in Andrographis Effervescent: Analyzed by HPLC with UV detection at 225 nm. The method must be validated as a Stability Indicating method to confirm separation from thermal degradation products.
• 6-Gingerol in Ginger Effervescent: Analyzed by HPLC with UV detection at 280 nm.
• All active ingredients must be maintained at Not Less Than 90% of the initial declared value throughout the stated shelf life. For vitamins and actives with known degradation rates under ASEAN Zone IVb conditions, manufacturers may include an Overage of 10 to 30% above the label-declared value in the formula to compensate for expected degradation during storage, ensuring label claims are met at the end of shelf life.

Long-term Stability testing uses 30°C / 75% RH conditions per ASEAN Zone IVb for a minimum of 24 months. Accelerated Stability testing uses 40°C / 75% RH for 6 months. For effervescent tablets that show unusual sensitivity to moisture in preliminary screening, Intermediate Stability testing at 30°C / 65% RH provides an additional data set.

• Standard Pull Points for Long-term testing are at months 0, 3, 6, 9, 12, 18, and 24. Standard Pull Points for Accelerated testing are at months 0, 1, 2, 3, and 6.
• Moisture Sorption Isotherm characterization using Dynamic Vapor Sorption (DVS) is conducted as a supplementary study to define the Critical Relative Humidity at which moisture uptake accelerates significantly. This data informs packaging specification selection and defines the maximum ambient humidity conditions for manufacturing operations.
• In-use Stability testing simulates the consumer experience of opening the tube or sachet and using tablets over days or weeks. For multi-tablet tube formats, samples are stored in open tubes under controlled temperature and humidity conditions at 25 to 30°C and assessed for Moisture Content, Effervescence Time, and active ingredient content at defined intervals. In-use Stability data is required for the label instruction “Close cap immediately after use” to be evidence-based.

Packaging design is the most critical factor in effervescent tablet stability, as it is the primary barrier against moisture ingress.

• Aluminium tubes sealed with Desiccant-containing Closures are the standard packaging format. The tube must have a Water Vapor Transmission Rate (WVTR) of less than 0.1 mg per day to provide effective moisture barrier. The closure system must incorporate a Built-in Desiccant such as Aquaguard or Desinet, containing Silica Gel or Molecular Sieve that continuously absorbs residual moisture in the headspace throughout shelf life.
• Sachet packaging uses Multi-layer Lamination construction with an Aluminium liner and a heat-sealable Polyolefin inner layer. The sachet size must be appropriate for single-use consumption to avoid In-use Stability concerns. Sachet construction must be verified for seal integrity by Burst Test and Leak Test before commercial launch.
• Strip Pack or Blister Pack using Aluminium-Aluminium Cold Form Foil provides the highest moisture barrier of any individual unit packaging format and is the preferred option for premium effervescent products with the highest moisture sensitivity.
• Desiccant systems used in aluminium tube closures include Silica Gel (effective for general moisture control), Molecular Sieve 4Å (effective for very low humidity control below 20% RH), Calcium Oxide (very high moisture absorption capacity, used for high-sensitivity formulations), and Calcium Sulfate. The appropriate desiccant type and quantity depends on the tube volume, number of tablets, and the Critical Moisture Content of the formulation.
• GMP manufacturing of effervescent tablets requires a controlled-humidity production environment at Relative Humidity of 25% or below, as specified in GMP guidelines for moisture-sensitive products. This requirement applies from granulation through compression, coating, and packaging operations.
• Product labels for effervescent tablets must include instructions: “Store in a dry place,” “Close cap tightly immediately after use,” and “Do not remove desiccant from tube,” as these instructions are essential to consumer compliance with moisture control requirements and form part of the stability evidence package for regulatory submission.
